Does anyone know if it is possible to run Rail Sim by creating a Virtual drive?

If your question relates to setting up a virtual drive in an attempt to use it as a replacement for a DVD drive with the RS disc in, then the answer is No.

The protection routine looks for the existance of the DVD in the drive.

I tried to install it on a USB flash drive to improve load times, but alas it wouldn't work. From memory it couldn't find files it needed. Maybe more time & persistance needed but didn't have either at the time. Not sure if this is what you are asking.
