I would suggest getting RWTools if you don't already have it, and check the scenario with that to see if you have all the required stock.

If you have everything that is required and RW works ok with other scenarios then the best thing you can do is contact AP customer services and ask their advice.

Have you had a look at http://www.armstrongpowerhouse.com/free ... Issues.pdf?

Did you do the usual trouble shooting step of clearing the games cache before loading the route/scenario?

The two 'missing marker' errors are of concern .. it may suggest a bad route installation? But, before you do a full cache verify:

Take gptech's advice and clear the cache on the Tools tab of the RW Launcher.

If that doesn't fix it, here's something else you can try:

1. Open the scenario in the Scenario Editor
2. Hit F2 and confirm the save
3. Hit the big arrow at the lower right of the RW window to go into play mode

No one seems to know why this works, but sometimes it sorts out problems with consists. If you still get the error, quit and restart RW and try opening the scenario as usual.

As a last ditch effort, you may have to do a full cache verify. This, of course, will overwrite any mods you've made to your Steam supplied routes, scenarios and stock, so it should be done only if absolutely necessary.

Good luck.

Yes, missing markers are a concern---if they actually are missing and not just a glitch related to this scenario you may find all other scenarios that reference them don't work.

How?... if you tell us what you did it would make helping you a lot easier, far better for us to give advice based on events rather than guessing at what may have happened.

Bit of a guess rather than actual knowledge/experience here, but have you used the feature in RW Tools to make all RW files read/write?
I'm thinking along the lines of a permissions conflict, where the update wants to re-write files but Windows own file protection kicks in and prevents it, or tries to prevent it, and you end up with some files being corrupted.

Verify the game files---might take more than 1 go though.
