So when 2x20s are on a nuclear flask run are they both powered? I'm assuming only one is powered and the other is there should the other fail.

It's not just the 20s, it's whatever traction is on the flasks.
66s and 57s only generally work within class, though 20s and 37s can be mixed. Unless they are an incompatible pairing, eg 37 and 66, then normally both locos are powering in multiple, with a crew member in the rear cab keeping an eye on the load.

Normally both locos power the train to save one loco doing the work and the other loco is already running should one of them shut down.
