I'm assuming that at some point, you had it working OK.

Try installing the latest Tsection file:

Click to view more informationMSTS Standardized global tsection.dat file Build 00042UK [325489 bytes] - Tsection42UK.zip
File ID: 13758 Date: 05 Jan 2012 - 25066 Downloads

Installing routes can often mean that your Tsection file is over-written by an older version, often causing MSTS to crash.

If that doesn't fix things, come back here and we'll think of something else.

Invariably the only message you get, if you get one at all, is that Train.exe has 'encountered a problem', so that part is not unusual.
my guess is that you have installed something new and that it has a fault of some kind. Use TrainStore to change to a different route and different activity, preferably one that does not use rolling stock common to the one that's crashing the sim, then try again.
If you still get a crash, try one of the default routes - the Orient Express is a good bet as that uses stock so totally different to UK stuff and almost nothing gets aliased to it. If that crashes, then you're probably into a reinstall as something fairly fundamental has gone pear-shaped.
If one of these options allows thing to start, then just narrow down the problem by trying activities for the questionable route one at a time until you find the one that crashes. You can try reinstalling it, or put yourself into diagnosis mode and see if you can figure out why it crashes.
