I´m writing an activity for the West Coast Express MSTS route, but I´m unsure if the London Midland semi-fast services from Birmingham/Crewe/Northampton to London Euston should operate over the slow lines all the way to Euston or if they change over to the fast lines after rejoining the mainline south of Northampton. Does anyone know?

Northampton - Euston semi-fasts, fast or slow lines?
Generally I think they use the slow lines, but I tend to travel Man - Eus at peak times when there is less freight and more Pendolino traffic. There may be more pathing on the fast lines off peak, 350s have got the acceleration to mix with the 390s - surprisingly this is as important as top speed.
If you have a look at WCML Gen group at yahoo, Chris Warman knows that patch and will be able to give you a more definitive answer.

I've seen up off-peak Northampton services routed onto the up fast at Ledburn Jct, but not consistently - depending on what was ahead on the up slow, I presumed. I'd be interested to know if there's a pattern.

As a regular traveller on these trains you will find that the trains that are Leighton Buzzard first or last stop use Ledburn Junction during the peaks as well as the off peaks.
